RTK’s
Receptor Tyrosine Kinases (RTK’s) zijn enzym-gekoppelde receptoren. Het zijn 2nd messengers. Een voorbeeld van een RTK is de insulinereceptor:
- Insuline bindt met monomere insulinereceptoren → er vindt dimerisatie van de receptoren plaats: ze komen bij elkaar
- Het intracellulaire deel van de receptor ondergaat autofosforylering: er komen een aantal gefosforyleerde tyrosines op de RTK: vormen bindingsplaatsen voor verschillende 2nd messenger eiwitten
- Eiwitten met een affiniteit voor fosfo-tyrosine binden aan de gesfosforyleerde tyrosines → er vormt een intracellulair signaal complex
- De eiwitten worden door fosforylering actief
Ras-eiwitten:
De meeste receptor tyrosine kinases activeren het monomere G-eiwit Ras. Ras-eiwitten fungeren als “moleculaire schakelaars” die intracellulaire signaalketens controleren:
- Een signaalmolecuul activeert receptor tyrosine kinases
- Een adaptor proteïne bindt aan de gefosforyleerde tryosines
- Ras-GEF (Guanine nucleotide Exchange Factor) bindt aan het aan het adaptor proteïne
- Ras-GEF zet GDP op het inactieve Ras-eiwit om in GTP → het Ras-eiwit wordt geactiveerd
- Geactiveerd Ras (met dus gefosforyleerde tyrosines) activeert een proteine kinase cascade, bijvoorbeeld de MAP-kinase route
- MAP = Mitogen Activated Protein Kinase (mitogeen = celdeling stimulerend)
- Een geactiveerd Ras bindt met een MAP-kinase die fosforyleert: dit wordt een MAP-kinase kinase kinase genoemd
- MAP-kinase kinase fosforyleert MAP-kinase kinase
- MAP-kinase kinase activeert MAP-kinase
- MAP-kinase stimuleert de celdeling door een aantal “target” eiwitten te fosforyleren
Ras activeert dus de MAP-kinase route
PI3-kinase:
Geactiveerde RTK’s stimuleren ook PI-3 kinase:
- PI3-kinase zet PIP2 om in PIP3
- PIP2 en PIP3 zijn eiwitten die met lipiden vast in het membraan zitten
- Door fosfotyrosine kan PI3-kinase binden en geactiveerd worden → forsoryleert PIP2 naar PIP3
- Aan PIP3 kunnen andere eiwitten, zoals PDK1, worden verbonden
- PDK1 bindt aan en activeert Akt
- Akt activeert Tor (ook een kinase)
- Tor stimuleert de celgroei via:
- Inhibering van de eiwitafbraak
- Stimulering van de eiwitsynthese
- Geactiveerd Akt remt de apoptose door de fosforylering/remming van het pro-apoptose eiwit Bad → dit zorgt voor het vrijgeven van Bcl2
- Bcl2 promoot de overleving van de cel
- Tor stimuleert de celgroei via:
Het effect van PI-3 kinase in omkeerbaar: PIP3 kan door PTEN omgezet worden in PIP2: PTEN haalt de fosfaatgroep van PIP3 af. Er is dus een balans tussen PIP3 en PIP2.
Kankercellen:
De meeste kankercellen hebben activerende mutaties in de RTK- en/of GPCR-signalering pathways die leiden tot:
- Celgroei
- Proliferatie
- Beweeglijkheid
- Invasie
- Overleving
Een voorbeeld hiervan is BRAF: een MAP-kinase kinase kinase. In 40% van de huidmelanomen zit er een mutatie in BRAF. Om dit mutant DNA aan te pakken is er een BRAF-remmer ontwikkeld: deze zorgt binnen enkele dagen voor een afname van het aantal melanomen. Echter is er na 4-8 maanden een terugval waarbij er sprake is van een aggresievere vorm. Tegenwoordig wordt dit opgelost door meerdere remmers te geven.
Cross-talk tussen GPCR- en RTK-signalering:
Er is veel cross-activatie van de verschillende routes, afhankelijk van de cellen: verschillende eiwitten kunnen dezelfde pathway activeren → PKC kan ook de MAP-kinase kinase kinase-pathways activeren. Daarnaast heeft een cel verschillende receptoren die de binnenkomende signalen integreren. De balans tussen de signalen en de hoeveelheid signalen bepaalt het effect.
Insulinesignalering
Na insulinestimulatie wordt in het celmembraan spiercellen en adipocyten het aantal glucosetransporters (GLUT4) verhoogd:
- Insuline bindt aan een receptor
- Een insulinereceptor (INSR) is een receptor tyrosine kinase →bestaat uit 2 α- en 2 β-ketens verbonden met difsulfide bruggen:
- α-ketens: bindt insuline
- β-ketens: bevat tyrosine kinase domeinen
- Er vindt dimerisatie van de receptoren plaats
- Het intracellulaire deel wordt geautofosforyleerd
- Een insuline receptor substraat (IRS) eiwit bindt en wordt d.m.v. fosforylering geactiveerd
- Er ontstaat een signaalcascade:
- Activatie van PI3-kinase
- Activatie van Akt
- Remming van AS160 (= Rab-GAP)
- Verhoging van Rab-GTP
- Glut4 vesicles (glucosetransporters) migreren naar en fuseren met het plasmamembraan
- Een hogere glucose inname is mogelijk
Doordat er constitutief, insuline onafhankelijk, glucosetransport door Glut2 plaatsvindt kunnen levercellen altijd glucose opnemen. Insulinereceptoren in het brein bepalen het eetgedrag: meer glucose = minder honger. Insuline speelt niet alleen een rol in de glucoseopname maar ook in de processen van andere cellen.
