De productie van mannelijke hormonen start met de vrijlating van gonadotropine vanuit de hypothalamus. De gonadotrope cellen zitten in de hypofyse en worden via het poortadersysteem bereikt. Wanneer de gonadotrope cellen worden geactiveerd gaan zij follikelstimulerend hormoon (FSH) en luteïniserend hormoon (LH) produceren. Luteïniserend hormoon zorgt voor de productie van testosteron in de Leydigcellen. De Leydigcellen zitten gepositioneerd in de testis en bevatten receptoren voor testosteron. Testosteron zorgt voor de stimulatie van Sertollicellen, het zorgt voor een toename in spiermassa en de ontwikkeling van secundaire geslachtskenmerken. Testosteron zorgt ook weer voor negatieve feedback op de secretie van gonadotropine en luteïniserend hormoon wanneer de productie hiervan voldoende is. De Sertollicellen worden gestimuleerd door follikelstimulerend hormoon en testosteron en ze zorgen voor de productie van voedingsstoffen en groeifactoren voor de maturatie van de spermacellen. Sertollicellen zorgen ook weer voor negatieve feedback op de secretie van follikelstimulerend hormoon wanneer de productie hiervan voldoende is door middel van inhibine.
